Rt-pcr Microdevice Integrated with a Rosgene Strip for Colorimetric Detection of Influenza H1n1 Virus

چکیده

A polymerase chain reaction (PCR) reactor was incorporated with an immunochromatographic ROSGENE strip for colorimetric detection of Influenza H1N1 virus. A reverse transcriptase-PCR (RT-PCR) cocktail, which contains Texas Redlabeled primers, biotin-dUTP, and Influenza H1N1 virus, was filled in the PCR chamber, and the target gene was amplified by RT-PCR. The resultant amplicons which were attached with biotin moieties and Texas Red haptens were first conjugated with the gold nanoparticle labeled antibody, and then captured on the streptavidin coated on the ROSGENE strip. The target virus is simply confirmed by observing a red color in the test line of the strip.
